The Hippo pathway is a highly conserved signaling pathway that plays crucial roles in regulating organ size, tissue homeostasis, and cell proliferation. It was initially discovered in Drosophila melanogaster and has since been found to be functionally conserved in mammals, including humans.

Key proteins and components of the Hippo pathway include:

1. **Mammalian STE20-like protein kinase 1/2 (MST1/2):** MST1 and MST2 are serine/threonine kinases that function as core kinases in the Hippo pathway. They form a complex with their regulatory protein Salvador (SAV1) and phosphorylate and activate the downstream kinase LATS1/2.

2. **Large tumor suppressor kinase 1/2 (LATS1/2):** LATS1 and LATS2 are serine/threonine kinases that are phosphorylated and activated by MST1/2. Activated LATS1/2, in turn, phosphorylates and inhibits the transcriptional co-activators Yes-associated protein (YAP) and WW domain-containing transcription regulator protein 1 (WWTR1, also known as TAZ).

3. **YAP and TAZ:** YAP and TAZ are transcriptional co-activators that regulate the expression of genes involved in cell proliferation, survival, and differentiation. When phosphorylated by LATS1/2, YAP and TAZ are sequestered in the cytoplasm, preventing their translocation to the nucleus and transcriptional activity. Conversely, when YAP and TAZ are dephosphorylated, they translocate to the nucleus and interact with transcription factors, such as TEA domain (TEAD) family members, to promote gene expression.

4. **TEAD transcription factors:** TEAD transcription factors are key downstream effectors of the Hippo pathway. They interact with YAP and TAZ in the nucleus to regulate the expression of genes involved in cell proliferation, survival, and tissue growth.

The Hippo pathway plays a critical role in controlling organ size and tissue growth by regulating cell proliferation, apoptosis, and stem cell maintenance. Dysregulation of the Hippo pathway has been implicated in various human diseases, including cancer, where aberrant activation of YAP and TAZ promotes uncontrolled cell proliferation and tumor growth.

Understanding the molecular mechanisms underlying the Hippo pathway and the roles of its key proteins in regulating cell growth and tissue homeostasis is essential for elucidating the pathogenesis of diseases associated with dysregulated cell proliferation and for developing therapeutic strategies to target the Hippo pathway for the treatment of cancer and other related disorders.
