Ferroptosis is a form of regulated cell death characterized by iron-dependent lipid peroxidation. Unlike apoptosis or necrosis, ferroptosis specifically involves the accumulation of lipid hydroperoxides, which ultimately leads to cell membrane damage and cell death.
One of the key proteins involved in regulating ferroptosis is glutathione peroxidase 4 (GPX4). GPX4 is an enzyme that plays a crucial role in protecting cells from oxidative damage by catalyzing the reduction of lipid hydroperoxides to their corresponding alcohols. In the context of ferroptosis, GPX4 acts as a critical antioxidant defense mechanism by scavenging lipid peroxides and preventing the accumulation of toxic lipid species.
However, inhibition or depletion of GPX4 results in the accumulation of lipid hydroperoxides, leading to ferroptotic cell death. This process can be triggered by various factors, including depletion of the antioxidant glutathione, inhibition of the cystine/glutamate antiporter system (system Xc-), or direct inhibition of GPX4 activity by small molecules such as RSL3.
Understanding the role of GPX4 and other key proteins in regulating ferroptosis is essential for elucidating the molecular mechanisms underlying this unique form of cell death and developing potential therapeutic interventions for diseases associated with dysregulated cell death processes.
